The event will see islanders make their way round a 13K course in Jersey.
It starts at 9am from People’s Park, St. Helier and finishing at Durrell Wildlife Park.
Film star Henry Cavill, one of Durrell's ambassadors, will be at the event.
A family fun day is also taking place at Durrell Wildlife Park after the race.
The weekend event coincides with International Pride Month and the 51st anniversary of the Stonewall riots.
The scheme has been backed by the island's Lieutenant-Governor, and is the first of its kind in the Channel Islands.
