Speculation had been building that a deal was imminent, and the 23-year-old has now signed a three-year deal with The Championship newcomers.
Gray scored 30 goals for the Hatters last season as they secured the Conference title.
"We are naturally disappointed to lose Andre but it goes without saying that we wish him well at Brentford," Luton manager John Still told the club's official website.
"With only a year left on his contract, it was either a case of him extending with us or him moving on. It is a great move for him and I am confident that he will go on and enjoy huge success at Griffin Park."
